Mobile Phase Preparation Documentation for Peptide Research Workflows

Documentation Structure and SOPs
Establish a version-controlled SOP that defines the minimum required documentation fields for every prepared mobile phase. Reagent and Batch Traceability
Traceability is critical for troubleshooting retention shifts, peak distortions, or reproducibility issues. Record supplier, catalog number, lot number, and Certificate of Analysis (COA) reference for each solvent and additive. For aqueous buffers, include water source (e.g., Milli-Q identifier), resistivity or conductivity grade as declared by lab policy, and the lot number of any salts used.
Assign a unique mobile phase batch ID and label the bottle/container with that ID, preparation date, and expiry or recommended use window established via stability studies. Retain a sample of each prepared batch when feasible so that later analytical testing can be performed if anomalies arise during peptide separations.
Data Integrity and Audit Trails
Ensure that all entries are attributable, legible, contemporaneous, original, and accurate (ALCOA principles). Prefer electronic records with secure audit trails: who entered the record, what was entered, when, and any subsequent edits with reasons. For paper-backed workflows, require ink signatures, witness signatures where appropriate, and scanning into the ELN within a defined timeframe.
Document instrument-ready checks that are relevant to mobile phases: pH meter calibration status (dates, buffers used, technician), balances calibration status, and any inline conductivity or pH readings measured after mixing. Record the identification of analytical instruments intended to use the mobile phase (instrument ID, method ID) to link mobile phase batches to chromatographic runs for post-run investigations.
Quality Control Checks and Acceptance Criteria
Define and document QC acceptance criteria that match the analytical risk profile: e.g., allowable pH range, measured conductivity, visible particulate acceptance, and degassing verification. Log measured QC values and the instrument or device used for measurement (including calibration status and last calibration date). Include a pass/fail decision record and the initials or electronic sign-off of the reviewer.
Maintain a simple incident log for deviations (e.g., out-of-specification pH, particulate observed after filtration) that ties the deviation to the batch ID, corrective action taken, and impact assessment on ongoing or archived chromatographic data. Link retention sample identifiers and failure investigations to chromatographic runs (sequence IDs) so that retrospective data integrity reviews are straightforward.
