ARA-290 Peptide in Preclinical Laboratory Research: Structure and Study Applications

Not for human consumption — research use only. This article is written for laboratory researchers and describes in-vitro research applications only.

Introduction

ARA-290 is an 11-amino-acid linear peptide with the sequence QEQLERALNSS, derived from the helix-B surface domain of human erythropoietin (EPO; UniProt P01588). It is studied in preclinical laboratory research as a non-hematopoietic EPO-derived peptide — a peptide designed from EPO’s structure but investigated in laboratory models separately from EPO’s classical red-blood-cell activity.

Erythropoietin itself is a 193-amino-acid glycoprotein cytokine whose classical receptor is the EPOR homodimer. Structure-guided dissection of EPO’s tertiary fold led researchers to ask whether discrete surface regions of the molecule could engage receptor complexes independently — the hypothesis that produced the helix-B-derived peptide series described by Brines and colleagues in the peer-reviewed literature.

This article reviews the molecular structure of ARA-290, the receptor systems investigated in laboratory research, and the analytical methods used to characterize reference material.

Molecular Structure

ARA-290 is a short linear peptide of eleven residues (Gln-Glu-Gln-Leu-Glu-Arg-Ala-Leu-Asn-Ser-Ser). Its sequence corresponds to a solvent-exposed region of the helix-B domain of erythropoietin, identified through structure-based analysis of EPO’s tertiary fold (Brines et al., Proc Natl Acad Sci USA 2008). At eleven residues, ARA-290 is readily accessible by standard Fmoc solid-phase peptide synthesis and presents a compact, linear epitope for receptor-interaction studies.

Structural features of interest to peptide chemists include the alternating charged and polar character of the sequence — glutamate and arginine residues flanking a hydrophobic leucine-alanine-leucine core — and the absence of cysteine residues, which simplifies synthesis and avoids disulfide heterogeneity in reference preparations.

From an analytical standpoint, the 11-residue length places ARA-290 in a favorable range for high-resolution mass spectrometry and complete MS/MS sequence coverage, while the absence of post-translational modifications keeps the theoretical mass calculation straightforward for identity confirmation.

Receptor Research in Laboratory Models

In the literature, ARA-290 and related EPO-derived peptides are studied in the context of a heteromeric receptor complex composed of the erythropoietin receptor (EPOR) and the common beta subunit CD131, described by Brines and Cerami (Mol Med 2012) as the innate repair receptor. Laboratory research models reported include:

  • Receptor-binding assays in cell systems expressing EPOR, CD131, or both, used to compare the binding profiles of EPO-derived peptides against the full-length EPO molecule.
  • Cell-based signaling readouts (for example, JAK-STAT pathway reporters) that distinguish EPOR/CD131-mediated signaling from classical EPOR-homodimer signaling in controlled in-vitro systems.
  • Preclinical laboratory models in which EPO-derived peptides serve as research tools for studying receptor pharmacology outside the hematopoietic system.

In these laboratory systems, ARA-290 functions as a pharmacological probe: by comparing its binding and signaling profile with that of native EPO and with scrambled-sequence control peptides, researchers map which receptor contacts are attributable to the helix-B region. Such comparative designs are standard in peptide–receptor interaction studies.

The “non-hematopoietic” designation used in the literature reflects laboratory findings that certain EPO-derived peptides engage the EPOR/CD131 complex without the red-blood-cell stimulating activity of native EPO — a distinction established through comparative assays rather than assumed from structure alone.

Analytical Characterization

Reference standards of ARA-290 are characterized with orthogonal methods:

  • Reversed-phase HPLC with UV detection provides the chromatographic purity profile. Short linear peptides such as ARA-290 typically resolve well on C18 columns with water/acetonitrile gradients containing TFA as an ion-pairing agent.
  • Mass spectrometry (ESI-MS) confirms the intact molecular mass against the theoretical value calculated from the QEQLERALNSS sequence.
  • Sequence verification by tandem MS (MS/MS fragmentation) or Edman degradation confirms the residue order — an essential check for a peptide whose behavior in receptor assays depends on the precise helix-B-derived sequence. The short length is advantageous here: complete b- and y-ion series are typically obtainable, allowing unambiguous confirmation of the residue order.
  • Amino acid analysis verifies bulk composition following acid hydrolysis.

COA documentation of HPLC purity, MS identity, and sequence confirmation precedes the use of any lot in receptor-binding or cell-based studies.

Lyophilized Handling

ARA-290 reference material is typically supplied as a lyophilized powder and stored at −20 °C or below in desiccated, light-protected conditions. Standard laboratory practice includes single-use aliquoting to avoid repeated freeze–thaw cycles and equilibration of sealed vials to room temperature before opening to prevent condensation. Because short linear peptides can be hygroscopic, vials are kept sealed with desiccant until use, and any prepared analytical stocks are tracked with preparation dates and storage temperatures in the laboratory’s sample-management records.

Not for human consumption — research use only. ARA-290 is supplied strictly as a laboratory reference material for in-vitro and preclinical laboratory research. It is not for human or veterinary use, and no medical, therapeutic, or dosing claims are made or implied in this article.
