Microscale Thermophoresis (MST) in Research Peptide Analysis: Binding Affinity and Interaction Studies

Microscale thermophoresis (MST) measures how molecules move along a microscopic temperature gradient, and how that movement changes when a binding partner is present. In research peptide analysis, MST is used in the laboratory to study binding affinity and molecular interactions in solution under controlled conditions.

How the method works: A capillary sample is locally heated with an infrared laser, and fluorescence is monitored as molecules migrate. A titration series produces a binding curve from which researchers estimate an equilibrium dissociation constant in vitro. Controls typically include buffer-only capillaries, a non-binding reference, and checks for aggregation or adsorption to capillary walls, either of which can distort the signal.

Applications and limits: MST uses small sample volumes and can work in complex buffers, which makes it useful for peptide interaction screening in research settings. Results depend on labeling strategy, buffer composition, and sample quality, so laboratories confirm key findings with an orthogonal method such as surface plasmon resonance or isothermal titration calorimetry, and verify peptide identity and purity by HPLC and mass spectrometry first.
