Transmission Electron Microscopy (TEM) in Research Peptide Analysis: Ultrastructure and Aggregation Imaging

Transmission electron microscopy (TEM) gives researchers direct images of peptide assemblies at nanometer resolution, complementing light-scattering and chromatographic methods.

What TEM shows: With negative staining or cryo-TEM, laboratories can visualize fibrillar structures, amorphous aggregates, and particle morphology that bulk methods only infer. Grid preparation, stain choice, and imaging conditions must be documented because preparation can itself alter assemblies.

Use in a QC workflow: TEM is typically paired with SEC, DLS, and HPLC so morphology findings are cross-checked against size distribution and purity data. Include buffer-only and reference controls, image multiple grid regions, and report scale and magnification with every micrograph.

Limitations: TEM samples a tiny fraction of a preparation; it supports, but does not replace, quantitative purity and identity testing by HPLC and mass spectrometry.

Disclaimer: For laboratory research purposes only. Not for human consumption. The methods described are analytical laboratory techniques; no medical or therapeutic claim is made.
